ΕΡΕΥΝΗΤΙΚΑ ΕΝΔΙΑΦΕΡΟΝΤΑ

H ερευνητική μου εμπειρία ξεκίνησε με επίκεντρο τη χημεία, ανάλυση, βιοχημεία, ανοσοχημεία και μοριακή βιολογία των λιπιδίων. Η εμπειρία αυτή αξιοποιήθηκε σε μελέτες της ανοσολογικής κατάστασης των κυττάρων του πνεύμονα και στη ρύθμιση του πνευμονικού επιφανειοδραστικού παράγοντα, τόσο στη φυσιολογική αναπνοή, όσο και σε ασθενείς με μηχανική υποστήριξη της αναπνοής. Οι μελέτες αυτές γίνονται σε κυτταρικές σειρές, σε πρωτογενείς καλλιέργειες και σε βρογχοκυψελιδικό έκπλυμα (BAL). Στο πλαίσιο αυτό ιδιαίτερα ασχοληθήκαμε με το ρόλο και ρύθμιση της έκφρασης φωσφολιπασών Α2, ενώ πρόσφατα διερευνούμε τον ρόλο των εξωκυττάριων κυστιδίων/εξωσωμάτων στη φλεγμονή του πνεύμονα, καθώς και σε μεταβολές του φορτίου τους σε συνάρτηση με ασθένειες του πνεύμονα.

Ως επέκταση της προηγούμενης εμπειρίας και ερευνητικών ενδιαφερόντων, ένας τομέας που μας απασχολεί είναι η βιοδραστικότητα ξενοβιοτικών ενώσεων που απομονώνονται φαρμακευτικά και αρωματικά φυτά και φυσικών προϊόντων, η ανάλυση και μοριακός χαρακτηρισμός αυτών,καθώς και οι σηματοδοτικές πορείες που επάγονται στα κύτταρα από τις ενώσεις αυτές. Ιδιαίτερα μας ενδιαφέρουν η κυτταροτοξική, αποπτωτική, (ιδίως στην περίπτωση επιλεκτικής δράσης σε καρκινικά κύτταρα έναντι φυσιολογικών), η αντιφλεγμονώδης, αντιοξειδωτική και αντιμικροβιακή δραστικότητα. Στην ίδια κατεύθυνση, και σε συνεργασία με άλλους ερευνητές, ασχολούμεθα με τον μεταβολισμό ξενοβιοτικών και τοξικών ενώσεων στο περιβάλλον.

Αναλυτικά, παρατίθενται τα ερευνητικά ενδιαφέροντα της ομάδας:

Ανάλυση λιπιδίων. Με GLC, HPLC, MS (FAB, EI,Orbitrap)

  • Βιοχημεία των λιπιδίων. Μεταβολικές πορείες. Ανάλυση και χαρακτηρισμός λιπιδίων και μεταβολιτών τους και προσδιορισμός ενζυμικών δράσεων. Φωσφολιπάσες: ανίχνευση, προσδιορισμός, μεταβολική δράση.  Παράγοντας Ενεργοποίησης των Αιμοπεταλίων (PAF).
  • Φωσφολιπάση Α2 : Βιοχημεία και Μοριακή Βιολογία
  • Βιοχημική και ανοσολογική μελέτη του βρογχοκυψελιδικού εκπλύματος (BAL) / Αντιφωσφολιπιδικά αντισώματα. Επιφανειοδραστικός παράγοντας των πνευμόνων (lung surfactant).
  • Φλεγμονή και απόπτωση στο πνευμονικό παρέγχυμα βαρέως πασχόντων: Ποιοτικές και ποσοτικές διαταραχές του επιφανειοδραστικού παράγοντα των πνευμόνων σε ασθενείς με πνευμονικές διαταραχές. Εξετάζεται η συμμετοχή λιπιδικών μεσολαβητών όπως του PAF καθώς και κυτταροκινών στην παθογένεση ασθενειών που έχουν ως αποτέλεσμα την καταστροφή του surfactant.

RESEARCH INTERESTS

My research experience is focused in lipid chemistry, analysis, biochemistry, immunochemistry and cell biology. Especially, this experience was deployed in studying the immune status of lung cells and lung surfactant regulation during physiological breathing and mechanical ventilation, in healthy and injured lung. In this context, we have studied extensively the physiological role and regulation of phospholipases A2 (PLA2s). We understood that certain functions of signalling events and especially exocytosis of PLA2s involve extracellular vesicles (EVs)/exosomes, which convey a modified cargo, depending on the external trigger (mechanical, chemical) or the disease (cancer, inflammation). In this regard, we focus on EVs/exososomes analysis, characterization and regulation.

Finally, we are interested in the biological activity of xenobiotics derived from aromatic and pharmaceutical plants and the signalling pathways they induce on lung cancer cells, in comparison with healthy cells.

  • Phosphatidylcholine analogs and Platelet-activating factor (PAF). Molecular characterisation, Identification and investigation of metabolic pathways (de novo, remodelling). Endogenous PAF inhibitors
